Description: Anti-SMAD4 (D351H) Mouse Monoclonal Antibody

Background: SMAD4 family member 4, alsoknown as SMAD4, is a protein that in human isencoded by the SMAD4 gene. It is often foundmutated in many cancers. It acts as a tumorsuppressor that functions in the regulation of the TGF-β signal transduction pathway, whichnegatively regulates growth ofepithelial cells and the extracellular matrix (ECM). SMAD4alterations have been found inmultiploid colorectal cancer and pancreaticcarcinoma . It is found inactivated in at least 50% of pancreatic cancers. It is also found mutated in the autosomal dominantdiseasejuvenilepolyposis syndrome (JPS).

Immunogen: A synthetic peptide from theinternal region of SMAD4 which includes the mutation of D351H, human origin.

Western blot:

Western blot analysis of recombinant SMAD4 (D351H) and wildtype proteins. PurifiedHis-tagged SMAD4 (D351H) protein (lane1) and corresponding wild-type protein (1-552aa, 65kd) (lane2) were blotted with anti-SMAD4 (D351H) monoclonal antibody (Cat. #26418).

Immunofluorescence:

Immunofluorescence of cells expressing SMAD4 proteins with anti-SMAD4 (D351H) antibody. HEK293T cells were transfected with pCDNA3-GFP-SMAD4 (D351H) plasmid ,pCDNA3-GFP-SMAD4 (WT) plasmid or pCDNA3-GFP-SMAD4 (D351N) plasmid, then fixed and stained with anti-SMAD4 (D351H) monoclonal antibody (Cat. #26418).
